چکیده

Metabolic DDT resistance in Drosophila melanogaster has previously been associated with constitutive over-transcription of cytochrome P450s. Increased P450 activity has also been associated with increased oxidative stress. In contrast, over-transcription of glutathione S transferases (GSTs) has been associated with resistance to oxidative stress. However, little is known in regards to the impact of xenobiotics on induction of P450s and GSTs and if there exist differences in inducibility between the pesticide susceptible and resistant strains. Thus, we investigated the transcriptional expression of GSTs and P450s in DDT resistant (Wisconsin) and susceptible (Canton-S) Drosophila strains in response to exposure to DDT and the oxidative stressor H2O2. Wisconsin constitutively over-transcribed P450s, constitutively under-transcribed 27% of its total GSTs, and was more susceptible to H2O2 than Canton-S. DDT exposure induced GST expression only in the Wisconsin strain and not in the Canton-S strain. These results suggest that there are potentially more differences between pesticide susceptible and resistant strains than just constitutive expression of P450s; there may also exist, at least in some strains, differences in their patterns of inducibility of P450s and GSTs. Within the context of the Wisconsin strain, these differences may be contributing to the fly lines increased susceptibility to oxidative stress. 2011 Elsevier Inc. All rights reserved.

A Glycine Insertion in the Estrogen-Related Receptor (ERR) Is Associated with Enhanced Expression of Three Cytochrome P450 Genes in Transgenic Drosophila melanogaster

Insecticide-resistant Drosophila melanogaster strains represent a resource for the discovery of the underlying molecular mechanisms of cytochrome P450 constitutive over-expression, even if some of these P450s are not directly involved in the resistance phenotype. For example, in select 4,4'-dichlorodiphenyltrichloroethane (DDT) resistant strains the glucocorticoid receptor-like (GR-like) potent...

Expression of Cyp6g1 and Cyp12d1 in DDT resistant and susceptible strains of Drosophila melanogaster.

The Rst(2)DDT locus (loci) in Drosophila is associated with the over-expression of two cytochrome P450 genes, Cyp6g1 and Cyp12d1. Using northern and western blot analysis we observed the expression pattern of these two genes in two DDT susceptible (Canton-S and 91-C) and three DDT resistant strains (Wisconsin, 91-R and Hikone-R). In Canton-S and 91-R, the CYP6G1 protein was constitutively expre...

Characterization of the promoters of Epsilon glutathione transferases in the mosquito Anopheles gambiae and their response to oxidative stress.

Epsilon class GSTs (glutathione transferases) are expressed at higher levels in Anopheles gambiae mosquitoes that are resistant to DDT [1,1,1-trichloro-2,2-bis-(p-chlorophenyl)ethane] than in insecticide-susceptible individuals. At least one of the eight Epsilon GSTs in this species, GSTe2, efficiently metabolizes DDT to DDE [1,1-dichloro-2,2-bis-(p-chlorophenyl)ethane]. In the present study, w...

Determination of Glutathione S-Transferase e2 Region (GSTe2) in DDT Susceptible and Resistant Anopheles stephensi Populations: Significance and Application of Nucleotide and Amino Acid Comparison

Glutathione S-transferases (GSTs) are a major family of detoxification enzymes which possess a wide range of substrate specificities. Interest in insect GSTs has primarily focused on their role in insecticide resistance. In this study, following World Health Organization (WHO) routine susceptibility test, DNA was extracted from specimens of Anopheles stephensi collected from the Kazeroon distri...

Transcriptomic profiles of Drosophila melanogaster third instar larval midgut and responses to oxidative stress.

Oligoarray analysis was used to determine the number and nature of genes expressed in third instar Drosophila melanogaster larval midguts. The majority of transcripts were associated with protein synthesis and metabolism. Serine proteases were the main proteolytic enzymes detected. Some 40% of the cytochrome P450 genes and 74% of the glutathione S transferases (GSTs) in the genome of D. melanog...
